In recent years, DNJ has been favored by scholars at home and abroad, and many countries have developed a variety of health foods containing Natural Mulberry Leaf Extract DNJ. The following review summarizes the isolation and extraction of DNJ from mulberry leaves, in order to promote the development and utilization of DNJ from mulberry leaves.

Water extraction is the primary method for DNJ extraction. Mulberry leaf DNJ has strong water solubility, and the traditional water extraction method can only extract free DNJ, which is not effective for binding DNJ. The method is simple and reproducible, but the extract has many water-soluble impurities, and the separation effect is not good.
DNJ belongs to alkaloid compounds and is easily soluble in acidic medium. Hot acid can soften plant cell walls, which is beneficial for DNJ extraction. Therefore, the yield of acid extraction is significantly higher than that of water extraction.
Alkaloids and their salts are soluble in organic solvents such as ethanol, methanol and acetone. Ethanol has good dissolving performance, strong penetrating power to plant cell wall, and can accelerate the leaching of alkaloids from mulberry leaves. Generally, dilute ethanol or acidic ethanol is used as the extraction solvent. The extraction solution of the method is not easy to be moldy and deteriorated, and the ethanol is less toxic, and can be recycled and reused. Therefore, the method of extraction with ethanol is one of the most commonly used methods in history.
The microwave/ultrasonic-assisted extraction method has the advantages of rapidity, simplicity, high yield, and less damage to effective components. It has good application prospects in extracting natural active ingredients. The method utilizes microwaves to promote the transition of the rotational ability of the medium, aggravates thermal motion, and the ultrasonic waves produce cavitation, mechanical disturbances and other physical effects to break the plant cell wall. Combined with appropriate extractant, DNJ leaching is accelerated, thereby increasing the yield.
